标题 甘草渣中黄酮提取物质量标准研究

摘要:目的 建立甘草渣中黄酮提取物的质量标准,为甘草渣中提取的总黄酮提供质量控制的方法及依据。方法 采用薄层色谱法将提取物与甘草苷及甘草对照药材对比鉴别,采用紫外分光光度法测定总黄酮含量,EDTA二钠滴定法测定钙离子含量。结果 供试品色谱中,在与对照药材色谱相应的位置上显相同颜色的斑点,在与对照品相应的位置上显相同颜色的荧光斑点。紫外分光光度法测定甘草苷在3.32~9.96 ?g/mL范围内线性关系良好(r=0.999 7),吸光度在0.3~0.7之间,平均加样回收率为102.41%(RSD=3.22%),3批甘草渣黄酮提取物中总黄酮平均含量为8.82%,钙离子平均含量为0.107%(RSD=3.61%)。结论 本方法重复性好、灵敏度高、结果准确,可用于甘草渣中黄酮提取物的质量控制。

Abstract: Objective To establish the quality standard of flavone extracts in licorice residue; To provide the methods and basis for the quality control of total flavonoids extracted from licorice residue. Methods TLC was applied to compare the extracts with liquiritin and licorice; UV-spectrophotometry was used to measure the content of total flavones; EDTA-2Na titration was used to determine the content of calciumion. Results In the chromatogram of the test product, the spots of the same color were displayed on the corresponding position of the control medicinal material, and the fluorescent spots of the same color were displayed on the corresponding position of the control product. UV-spectrophotometry showed glycyrrhizin had a good linear relationship in the range of 3.32–9.96 ?g/mL (r=0.999 7), the absorbance between 0.3 and 0.7. The average recovery rate was 102.41% (RSD=3.22%), and the average content of total flavonoids in three batches of flavonoids extract was 8.82%. The average content of calciumion was 0.107% (RSD=3.61%). Conclusion This method has good repeatability, high sensitivity and accurate results, and can be used for the quality control of flavonoids in licorice residue.

甘草為豆科甘草属甘草Glycyrrhiza uralensis Fisch、胀果甘草Glycyrrhiza inflata Bat.、光果甘草Glycyrrhiza glabra L.的干燥根和根茎,有效成分复杂,主要为甘草酸和黄酮类化合物[1],但目前开发、生产的主要是甘草中的甘草酸[2],剩余物作为药渣弃去。研究表明,甘草渣中富含黄酮类化合物[3],具有多种生物活性,除消炎、抗菌、抗变态反应等,在抗氧化、抗癌、防癌、防肿瘤及保肝护消化道等方面作用显著[4]。
近年来,随着对甘草药理作用的进一步认识,全球对甘草的需求日益增长,野生甘草资源急剧减少,与此同时,甘草提取甘草酸或甘草浸膏后剩余的残渣(甘草渣)也逐渐成为危害生态环境的又一物质。本课题前期进行了“基于化学转化法破壁富集甘草药渣中总黄酮的开发研究”(兰州市科技局,2014-2-32)。为了保证甘草渣黄酮提取物的质量稳定性,本研究参考2015年版《中华人民共和国药典》(以下简称《中国药典》)甘草项进行甘草渣黄酮提取物的研究,建立其薄层色谱鉴别法、总黄酮含量测定法及钙离子含量测定方法等质量标准,以期为甘草渣中黄酮资源的有效、合理开发和质量评价提供依据。
